T4 DNA Ligase can be used to join DNA fragments with staggered or blunt ends and to repair nicks in double-stranded DNA having 3'-hydroxyl and 5'-phosphate ends. The enzyme is isolated from E. coli lambda lysogen.
T4 RNA ligase catalyzes the ATPdependent covalent joining of a 5´-phosphorylterminated nucleic acid donor to a 3´-hydroxylterminated nucleic acid acceptor. Substrates are single stranded DNA and RNA as well as mononucleosides 3´, 5´-biphosphates (pNp) (1). An oligonucleotide which has both a 5´-phosphate and a 3´-hydroxyl can serve as well as either acceptor as an donor, yielding either circular or multimeric products.

dnaB helicase is an enzyme in bacteria which opens the replication fork during DNA replication. Initially when DnaB binds to dnaA, it is associated with dnaC, a negative regulator. After DnaC dissociates, DnaB binds dnaG.The DnaB helicase is the product of the dnaB gene. The helicase enzyme that is produced is a hexamer in E. coli, as well as in many other bacteria. The energy for DnaB activity is provided by NTP hydrolysis. Mechanical energy moves the DnaB into the replication fork, physically splitting it in half.
