Ciramadol is an opioid analgesic that was developed in the late 1970s and is related to tramadol, tapentadol and venlafaxine. It is a mixed agonist-antagonist for the μ-opioid receptor with relatively low abuse potential and a ceiling on respiratory depression which makes it a relatively safe drug. It has a slightly higher potency and effectiveness as an analgesic than codeine, but is weaker than morphine. Other side effects include sedation and nausea but these are generally less severe than with other similar drugs.
Cisapride is a gastroprokinetic agent, a drug which increases motility in the upper gastrointestinal tract. It acts as a serotonin 5-HT4 receptor agonist and indirectly as a parasympathomimetic. Stimulation of the serotonin receptors increases acetylcholine release in the enteric nervous system. It was discovered by Janssen Pharmaceutica in 1980. In many countries it has been either withdrawn from the market or had its indications limited because of side effects.

Clonidine is a medication used to treat several medical conditions. It is a direct-acting α2 adrenergic agonist and an imidazoline. It has been prescribed historically as an antihypertensive drug. It is becoming a more accepted treatment for insomnia, as well as for relief of menopausal symptoms. It is increasingly used in conjunction with stimulants to treat attention-deficit hyperactivity disorder, for which it is administered in late afternoon or evening for sleep, and because it sometimes helps moderate ADHD-associated impulsive and oppositional behavior, and may reduce tics, a problem in which a part of the body moves repeatedly, quickly, suddenly and uncontrollably. Clonidine can be used in the treatment of Tourette syndrome. Its epidural use for pain during heart attack, postoperative and intractable pain has also been studied extensively. It has also been used to treat the hyperarousal symptoms of PTSD. It is also a mild sedative, and can be used as premedication before surgery or procedures. This medication may also be used to ease withdrawal symptoms associated with the long-term use of narcotics, alcohol and nicotine. In addition, clonidine has also been used for migraine headaches and hot flashes associated with menopause.
